Induction of Autocrine Growth Factor Activity Phenotype of Human Neuroblastoma Cells: Evidence for Expression Advances the Malignant MYCN
نویسندگان
چکیده
Amplification and enhanced expression of the MYCN oncogene are thought to contribute to the development and progression of human neuroblastomas. Here, we have transfected human neuroblastoma cells that harbor a single MYCN gene copy with the human MYCN gene driven by a viral enhancer/promoter, and we have compared the properties of the parental and the transfected cells. The transfected cells show an enhanced expression of the exogenous MYCN gene. Unlike the parental cells, they have acquired an increased proliferative potential, induce tumors in nude mice, grow in soft agar, and require low amounts of exogenous growth factors in order to proliferate. The A/yC/V-transfected, but not the parental, cells can synthesize and utilize autocrine growth factor activity. These results demonstrate that enhanced MYCN expres sion contributes to malignant progression of human neuroblastoma cells, conceivably by stimulating the expression of autocrine growth factor activity.

Neuroblastoma is the most common pediatric cancer in infants, arising from the sympathoadrenal lineage of the neural crest. Despite recent advances in other pediatric cancers, long term survival in high risk cases of neuroblastoma remains below 40%. Therefore, to develop successful therapeutics targeting high risk tumors, further research into the mechanisms involved in high risk tumor formatio...

Neuroblastoma is the most frequent extracranial solid malignancy of childhood with a high mortality in advanced tumour stages. The hallmark of neuroblastoma is its clinical and biological heterogeneity. The molecular mechanisms leading to favourable or unfavourable tumour behaviour are still speculative. However, amplification of the oncogene MYCN and expression of the neurotrophin receptor Trk...

Neuroblastoma, one of the common malignant childhood tumors, arises from neuroblast cells derived from theneural crest and destined for the adrenal medulla and the sympathetic nervous system and shows remarkable biologicalheterogeneity, resulting in favorable or unfavorable outcomes. Some tumors make rapid progress with a fataloutcome. In other instances, the tumors regress spontaneously in inf...
